Recognizing Mariano's spiritual calling, he was ordained a priest of the archdiocese of Sevilla on 22 December 1923.

After his ordination, Father Mariano started serving as an assistant pastor in Huelva on 27 April 1934. He quickly gained a reputation for his compassionate nature and his unwavering dedication to his parishioners. However, the onset of the Spanish Civil War brought about great turmoil and persecution for Catholics in Spain.

Tragically, Father Mariano's parish church was targeted by anti-Catholic forces on 21 July 1936, resulting in its destruction. Realizing the imminent danger, he went into hiding along with some of his devoted parishioners, seeking refuge from the violent upheaval. Despite their efforts to conceal themselves, Father Mariano was eventually discovered by his persecutors.

On his way to prison, Father Mariano was shot in the back, suffering a severe injury. He succumbed to his wounds soon after, passing away on 23 July 1936 in a hospital in Huelva due to blood loss.
